Biotium HL-60 Antigen / Human Leukemia Antigen (IPO-M6), CF740 conjugate, 0.1mg/mL
This antibody reacts with human leukemia cell line HL-60 and immunoprecipitates two proteins with MW of 48 kDa and 52 kDa. It does not stain B-cell lines Daudi, PHS, Namalwa, RPMI-1788 and T-cell lines CCRF-HSB2, Jurkat and Molt-4. This antibody stains monocytes and up to 10% of lymphocytes from peripheral blood of healthy donors. Blast cells of patients with AMMonL (M5 following FAB classification), AMMonL (M4) and hairy cells leukemia are positive. Its antigen is particularly expressed on blood cells from patients with infectious mononucleosis and CLL. Histiocytes and macrophages are also positive. Malignant cells from patients with AML (M1 and M2), T-ALL, B-ALL are not stained. HL-60 cells are used as an in vitro model of acute promyelocytic leukaemia and for differentiation and apoptosis studies. This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. This antibody recognizes a protein of 30-33 kDa, which is identified as CD20. Its epitope is located in the cytoplasmic domain of CD20 and was, therefore, ascribed as CD20cy in the 5th Workshop. CD20 is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. HHV 8 encodes a latent nuclear antigen (LNA), which is the product of the viral gene orf 73. LNA is capable of forming a complex with retinoblastoma susceptibility gene product, which may be related to its oncogenic activity. HHV8 is associated with three different diseases observed in AIDS patients; kaposi's sarcoma, primary effusion lymphoma (which is a rare type of non-Hodgkin lymphoma affecting the body cavities) and multicentric Castleman's disease. Reacts with a protein of 57 kDa, identified as the L1 protein of human papilloma virus type 16 (HPV-16). It is the major capsid protein of HPV-16.
